PathwayType: signaling Organ: generic Pathway_Author: A. Nesterova ORCID:0000-0002-9448-8101 Organ_System: generic Description: In humans, the genome is packaged into 46 pairs of sister chromatids during mitosis. Pathway is built manually using published studies. Tissue: generic Link: https://mammal-profservices.pathwaystudio.com/app/sd?urn=urn:agi-pathway:uuid-63c6e3a2-a695-46dc-a621-be77b09e923e NodeType: Pathway Notes: Headnote: In humans, the genome is packaged into 46 pairs of sister chromatids. The accurate partition of the genome during mitosis occurs owing to proper chromosome condensation and cohesion. Signaling description: DNA topoisomerase (ATP-hydrolyzing), condensing, and cohesin complexes are essential to these processes. The cohesin complex is critical for sister chromatid cohesion, while condensin and topoisomerase II are the major regulators of chromosome condensation. The mechanism of cohesin binding to DNA may share similarities with DNA binding of condensing. Namely, both complexes consist of heterodimers of SMC (structural maintenance of chromosomes) subunits which form gigantic tripartite ring structures. Condensin acts as an intramolecular crosslinker by grabbing sites on a single DNA strand and bringing them together, whereas cohesin acts as an intermolecular crosslinker by grabbing and holding two different sister chromatids. Topoisomerase II interacts with two DNA strands to effect strand passage. Outcome effects: The association between topoisomerase II and CDK1 likely represents the earliest step in mitotic chromatin condensation. Source: Cell Process CellType: generic |
