DEFINITION

Apolipoprotein(a) (4548 aa, ~501 kDa) is encoded by the human LPA gene. This protein is involved in the regulation of thrombosis and lipid metabolism.

DesignNote

Lipoprotein(a) (Lp(a)) is formed by the disulfide linkage of apolipoprotein B100 (APOB) of a low-density lipoprotein particle to apolipoprotein(a). (OMIM)

Apolipoprotein(a) (apo(a)) is proteolytically cleaved, to form mini-Lp(a). These apo(a) fragments may accumulate in atherosclerotic lesions and promote thrombogenesis. O-glycosylation of apo(a) may limit proteolytic fragmentation. The shared homology of mini-Lp(a) with plasminogen kringles IV and V is thought to underlie the atherogenicity of the protein, as the fragments may compete with plasminogen for fibrin(ogen) binding.
