FKBP12-Rapamycin Complex Associated Protein 1 contains 8 HEAT repeats and is a target for the G1 cell-cycle arrest and immunosuppressive effects of FKBP12-rapamycin complex. mTOR regulates ribosome biogenesis and cell growth in response to ATP or nutrient concentrations. FRAP phosphorylation of p53 (inhibited by rapamycin) downregulates PP2A, activates BAX, and induces mitochondrial membrane permeability and caspase activation. Mitogen-induced phospholipase D-dependent phosphatidic acid interacts with the mTOR rapamycin target domain. Cellular stress inhibits MTOR activity by stabilizing MTOR-RAPTOR binding, which also promotes MTOR phosphorylation of 4EBP1 and enhances MTOR activity toward S6K1. MTOR binds UBQLN1. |
