A serine/threonine protein kinase involved in cell cycle control, Cdk4 may complex with D-type G1 Cyclins and belongs to the CDC2/CDKX subfamily. CDK4/CDK6 phosphorylates Rb in early G1, displacing histone deacetylase and blocking transcriptional repression. CDK4 is inhibited by p16 (Cyclin-Dependent Kinase Inhibitor-2).
